GFP Expression in Mz1369: Frontal Projections

Sagittal Projections | Horizontal Projections

Eight hardware blended projections, proceeding from anterior to posterior in the hemisphere, were made with the extended focus algorithm of Bitplane Imaris.

Distal is to the top and proximal to the bottom in this series.
Red is mab22C10 fluorescence and green is GFP-fluorescence.

The optic stalk is seen entering the brain hemisphere. It is ensheathed by glial cells expressing GFP. Cells of the outer optic anlage, which produce neurons of the medulla and lamina, also express GFP, but at a lower level than in the inner optic anlage. The arrow labels the furrow that separates the region of the ooa that gives rise to the lamina from that which gives rise to the medulla.

This section cuts into the hemisphere and shows the photoreceptor axons spreading out. Cell body fibres that project from the medulla cortex to the medulla neuropil can be seen.

This section cuts into the medulla neuropil. Regularly arranged GFP expressing terminals of photoreceptors are visible. The developing lamina cortex is also targeted by GFP expression. 22C10 labelling highlights two bundles of T&C cells. Cell body fibres can be followed from the medulla cortex to the transient medulla neuropil, which is stained strongly by 22C10. The actual medulla neuropil itself is hardly labelled by 22C10.

This plane of section cuts the medulla open and reveals the lobula complex. T&C cell bundles can be followed as they run along the inner face of the medulla neuropil. The inner optic anlage becomes visible at the base of the medulla.

The curved medulla neuropil is cut into two halves in this section. Projections of the inner chiasm connecting medulla and lobula complex can be followed. Strands of cells proliferated from the inner optic anlagen between medulla and lobula are indicated by arrows.

The series leaves the neuropils of the optic lobe. The two limbs of the inner optic anlage are visible at the base of the optic lobe.
