Figure 1
Expression of the EXD and HTH Proteins in the Developing Brain.

(A-D) Expression of the EXD and HTH proteins at mid-stage 12. Embryos were labeled with anti-EXD (A, orange; Mann and Abu-Sharr, 1996) or anti-HTH (B-D, orange; Kurant et al., 1998) in conjunction with a neuron-specific anti-HRP (green) antibody. (A-C) Lateral image. (D) Dorsal image. Stars indicate neuroblasts. In C and D, embryos also were labeled with an anti-Fas II antibody (blue; Grenningloh et al., 1991). The identified fiber tract founder clusters are labeled according to Nassif et al. (1998). EXD also is expressed in the optic lobe primordia (ol). (E, F) Expression of the EXD (E) and HTH (F) proteins at stage 14. Dorsal images. Arrows, EXD/HTH cells in neuromere b1; arrowheads, EXD/HTH in neuromere b2; open arrows, EXD/HTH cells in neuromere b3. Reconstruction of optical sections. Embryonic anterior is to the left. See Hirth et al. (1995) for neuromeric organization of the embryonic brain. (G) Expression of the EXD protein in hth mutant. Dorsal image of laser confocal microscopy of stage 14 embryo double labeled with anti-EXD antibody (orange) and neuron-specific anti-HRP antibody (green). The brain commissure is not included in this reconstruction. (H) Expression of the HTH protein in maternal and zygotic exd null mutant embryo. Dorsal image of laser confocal microscopy of stage 15 embryo double labeled with anti-HTH antibody (orange) and neuron-specific anti-HRP antibody (green). (I) Fas II patterns in the developing brain at stage 14. Same embryo as in F and J but shows only the Fas II immunoreactivity. D/T, P1, P2l, P2m, P3l, P3m, P4l, and P4m indicate fiber tract founder clusters according to Nassif et al. (1998); MT, medial cervical tract; LT, lateral cervical tract; LPT, lateral protocerebral tract. The D/T cluster consists of two distinct clusters at this stage belonging to the posterior parts of neuromere b2 and b3. (J-L) Expression of the HTH and FAS II proteins at stage 14. Same embryo as in F showing HTH (red/pick) and FAS II (blue/pink). (K) A close up image of the upper part of J, reconstructed with dorsal sections. (L) Lateral image. Reconstructions of medial optical sections. Note the co-expression of the HTH and FAS II proteins in many of the fiber tract founder clusters. The HTH protein is localized in nuclei whereas FAS II is localized on the cell surface and pronounced in axons as axogenesis proceeds. MPT, medial protocerebral tract; fg, frontal ganglion of the stomatogastric nervous system. Scale bar, 20 µm; (A, E, I), 10 µm; (D, K, L).
